以化合物(CAS: 131543-46-9)和正丁醛為原料合成2-丙基咪唑的一般步驟:向NH4HCO3(16.45 g,208.1 mmol)的水(10 mL)懸浮液中依次加入正丁醛(9.2 mL,7.52 g,104 mmol)和40% w/w乙二醛水溶液(11.9 mL,15.09 g,104.0 mmol)。將反應(yīng)混合物在室溫下攪拌過夜,隨后通過減壓蒸餾去除揮發(fā)性組分。用四氫呋喃(THF)萃取殘余物,合并有機(jī)相后再次減壓蒸餾,得到粗產(chǎn)物(11 g,收率96%)。粗產(chǎn)物通過柱色譜法純化(洗脫劑:二氯甲烷→甲醇/二氯甲烷,1:60→1:30),得到2-丙基咪唑(7.45 g,收率65%)。產(chǎn)物結(jié)構(gòu)經(jīng)1H NMR(500 MHz, CDCl3)和13C NMR(125 MHz, CDCl3)確認(rèn):1H NMR δ 11.50(s, 1H), 6.96(s, 2H), 2.72(t, J = 7.4 Hz, 2H), 1.77(sext, J = 7.4 Hz, 2H), 0.98(t, J = 7.4 Hz, 3H); 13C NMR δ 149.1, 121.4, 30.7, 22.3, 14.0。
