以2-氰基噻唑為原料合成2-噻唑甲脒鹽酸鹽的一般步驟:在SQL反應(yīng)釜中加入甲醇(12L),隨后加入2-氰基噻唑(2200g,20.00mol,1當(dāng)量)和甲醇鈉(54g,1mol,0.05當(dāng)量)。將混合物在10℃下攪拌0.5小時。通過TLC(石油醚:乙酸乙酯 = 2:1)監(jiān)測反應(yīng)進(jìn)程,確認(rèn)原料消失并檢測到中間體的生成。接著,加入氯化銨(1296g,24.00mol,1.2當(dāng)量),并將混合物在65℃下攪拌16小時。再次通過TLC(石油醚:乙酸乙酯 = 2:1)確認(rèn)中間體消失,表明反應(yīng)完成。反應(yīng)混合物經(jīng)過濾除去固體氯化銨(210g),濾液經(jīng)干燥得到粗產(chǎn)物。粗產(chǎn)物用乙酸乙酯(20L)漿化2小時后過濾,得到2-噻唑甲脒鹽酸鹽(3200g),純度為98%。通過定量NMR測定,其有效含量為95%。分離收率為93%。1H NMR(400MHz,DMSO-d6)δ 8.26(d,J = 2.76Hz,1H),8.38(d,J = 3.01Hz,1H),9.81(br.s.,1H)。LCMS:m/z 127.9 [M + H]+。
