以1-(3-苯基氨基苯基)-乙酮為原料合成2-乙?;脏玎旱囊话悴襟E如下:首先,將苯胺與間乙酰基苯酚以1.2:1的質(zhì)量比加入反應(yīng)容器中,加熱至溶解。隨后,加入三氟甲磺酸(加入量為間乙?;椒淤|(zhì)量的0.025倍),在190℃下進(jìn)行脫水反應(yīng)。反應(yīng)過程中分離生成的水分。反應(yīng)6小時后終止。通過減壓蒸餾去除過量的苯胺,得到暗紅色固體,即3-乙?;桨罚ɑ衔顰)。接下來,將化合物A與硫(化合物A與硫的摩爾比為1:2.3)及250毫升丙酮(硫與單質(zhì)的總質(zhì)量為1摩爾)加入反應(yīng)容器中,加熱溶解形成透明液體。然后,加入碘(加入量為化合物A質(zhì)量的0.0125倍),在170℃下攪拌回流反應(yīng)。25分鐘后,反應(yīng)基本完成,停止攪拌,逐漸冷卻至120℃,此時可見黃色和黑色分層。小心傾出上層黃色液體,室溫冷卻后析出黃色固體。粗產(chǎn)物通過乙醇-水混合溶劑重結(jié)晶,得到淡黃色固體,即目標(biāo)產(chǎn)物2-乙?;脏玎?。本實(shí)施例的總收率為90.5%,HPLC純度為99.7%。

Nox1-dependent ROS generation has been shown to play a pivotal role in cell signaling, cell growth, angiogenesis, motility and blood pressure regulation. ML171 strongly blocks ROS generation in HT29 cells (IC 50 =0.129 μM) and only increasing over-expression of Nox1 can overcome the blockage of ROS generation caused by ML171 treatment in HEK293 cell system reconstituted with all the components required Nox1-dependent ROS generation. ML171 efficiently blocks ROS production measured by carboxy-H2-DCFDA staining as well as DPI used as a positive control. When ML171 is tested in HEK293-Nox1 reconstituted cell system, higher potency in blocking Nox1-dependent ROS generation is observed compared with the parental compound.
