以1,3,6,8-芘四磺酸四鈉鹽為原料合成溶劑綠7的一般步驟如下:在2000毫升三頸燒瓶中加入1200克濃硫酸,攪拌下加入120克1,3,6,8-芘四磺酸四鈉鹽,輕微加熱以促進(jìn)完全溶解后,緩慢升溫至90°C并維持1小時(shí)。反應(yīng)完成后,用水浴將反應(yīng)混合物冷卻至40°C以下,緩慢滴加480克液體硫磺黑,滴加過(guò)程控制在3小時(shí)內(nèi)完成。隨后,將反應(yīng)溫度維持在100°C左右繼續(xù)反應(yīng)5小時(shí)。反應(yīng)結(jié)束后,冷卻至室溫,將反應(yīng)混合物倒入720克冰水中,得到粘稠物料,經(jīng)過(guò)濾后干燥濾餅,得到芘四磺酸四鈉鹽。接下來(lái),將芘四磺酸四鈉鹽溶于1000毫升水中,轉(zhuǎn)移至2000毫升三口燒瓶中,用氫氧化鈉溶液調(diào)節(jié)至中性,再加入300克片狀氫氧化鈉。緩慢加熱至微沸狀態(tài),保持5小時(shí)。反應(yīng)完成后,冷卻,先用鹽酸調(diào)節(jié)pH值至7左右,然后加入100克氯化鈉攪拌至完全溶解,緩慢沉淀出淡黃色固體。經(jīng)過(guò)濾、干燥,最終得到約245克8-羥基-1,3,6-芘三鈉磺酸鹽。

The pH is calculated by measuring the ratio of fluorescent intensity at two excitation wavelengths, I463/I404, a measurement which is dependent on pH but independent of the concentration of the fluorophore and other variables which can alter the intensity. In the rabbit eye, Pyranine in the cornea and anterior chamber is observed to undergo easily measurable changes in fluorescent ratios associated with lid closure and contact lens wear, indicating its sensitivity to mild changes in pH.
