在-5℃條件下,通過(guò)注射器將亞硫酰氯(2.68g,0.023mol)緩慢滴加至甲醇(12mL)中,確保反應(yīng)溫度不超過(guò)-4℃。滴加完畢后,將反應(yīng)混合物在-5℃或更低的溫度下持續(xù)攪拌1小時(shí)。隨后,一次性加入4-氨基-3-(三氟甲氧基)苯甲酸(0.5g,0.003mol)[Alfa Aesar]。將反應(yīng)混合物轉(zhuǎn)移至環(huán)境溫度下攪拌3天。反應(yīng)完成后,通過(guò)真空濃縮去除溶劑。向殘余物中加入水(10mL),并一次性加入碳酸氫鈉(0.46g,5.4mmol),在環(huán)境溫度下攪拌30分鐘。使用乙酸乙酯對(duì)混合物進(jìn)行多次萃取。合并有機(jī)相,用硫酸鈉干燥,過(guò)濾后真空濃縮,得到4-氨基-3-(三氟甲氧基)苯甲酸甲酯1.02g(產(chǎn)率96.2%),初為紅色油狀物,冷卻后逐漸轉(zhuǎn)變?yōu)樽厣腆w。質(zhì)譜(MS):m/z 236.1(MH+)。1H NMR(500MHz,DMSO-d6)數(shù)據(jù):δ3.771(s,3H),6.329(s,2H),6.834(d,1H),7.6181(s,1H),7.655(dd,1H)。
