通用方法:通過4,6-二取代嘧啶-2-胺(16)的重氮鹽與濃鹽酸和ZnCl?反應(yīng)制備2-氯-4,6-二取代嘧啶(17)[35]?;衔?8的合成參考了文獻(xiàn)[32]的方法并進(jìn)行了改進(jìn)。向攪拌的哌嗪(45mmol)和K?CO?(16.5mmol)的水(20mL)溶液中,在50-65℃下分批加入2-氯-4,6-二取代嘧啶(17)(18mmol)。將反應(yīng)混合物在60-65℃下攪拌1小時(shí),隨后冷卻至35℃。過濾除去固體副產(chǎn)物1,4-雙嘧啶基哌嗪,濾液用氯仿萃取三次,有機(jī)相用Na?SO?干燥,減壓濃縮,得到化合物18,無需進(jìn)一步純化即可用于后續(xù)反應(yīng)。18a:黃色油狀物,產(chǎn)率88%。
