以2-溴-5-氟苯甲醚和氰化亞銅為原料合成2-甲氧基-4-氟苯腈的一般步驟:向含有1-溴-4-氟-2-甲氧基苯(9.0g)的N-甲基吡咯烷酮(100mL,Sure Seal; Aldrich)溶液中加入氰化亞銅(6.6g,73.7mmol,1.8當(dāng)量; Aldrich)。將反應(yīng)混合物在180℃下,于無水氮?dú)夥諊袛嚢?.5小時。反應(yīng)完成后,冷卻至室溫,加入14%的氨水溶液(330mL),繼續(xù)在室溫下攪拌45分鐘。用乙醚(100mL×3)萃取反應(yīng)混合物,合并有機(jī)相,依次用稀氨水、稀鹽酸和鹽水洗滌,然后用無水硫酸鎂干燥,濃縮后得到2-甲氧基-4-氟苯腈(5.2g,兩步收率85%)。產(chǎn)物為白色固體,其1H NMR(CDCl3,500MHz)數(shù)據(jù)如下:δ 3.91(3H,s,OMe),6.69(1H,dd,J = 2.3Hz,J = 10.5Hz,Ar-H),6.72(1H,dt,J = 2.5Hz,J = 8.0Hz,Ar-H),7.55(1H,dd,J = 6.5Hz,J = 8.5Hz,Ar-H);13C NMR(CDCl3,125.8Hz)數(shù)據(jù)如下:δ 56.49, 98.16, 100.06, 100.27, 108.31, 108.50, 115.83, 135.37, 135.46, 163.25, 163.34, 165.47, 167.50。通過乙醚研磨獲得分析樣品。元素分析計算值(C8H6FNO):C,63.57; H,4.00; N,9.26;實(shí)測值:C,63.36; H,3.91; N,9.16。
