一般步驟:在氮氣保護下,將0.99 M的乙基氯化鎂(EtMgCl,0.61 mL,0.6 mmol)的四氫呋喃(THF)溶液冷卻至0°C,緩慢滴加二環(huán)己胺(0.01 mL,0.05 mmol)和2-甲基噻吩(1a,0.048 mL,0.50 mmol)。將反應混合物升溫至60°C并攪拌24小時。隨后,依次加入1.4 mL THF和N,N-二甲基甲酰胺(DMF,0.5 mL,6.46 mmol),繼續(xù)在60°C下攪拌1小時。反應完成后,用飽和氯化銨水溶液(1.0 mL)淬滅反應,將混合物倒入乙醚/水的混合溶劑中,分離有機相和水相。水相用乙醚萃取兩次,合并有機層并用無水硫酸鈉干燥。減壓濃縮有機相得到粗產(chǎn)物,通過硅膠柱色譜法純化(洗脫劑:己烷/乙酸乙酯=20/1),得到5-甲基-2-噻吩甲醛(1a-CHO,62.3 mg,無色油狀物,收率99%)。
