名稱	Epirubicin hydrochloride
描述	Epirubicin hydrochloride (Pharmorubicin) is an adriamycin derivative, a topoisomerase (Topo) inhibitor, and a Forkhead box protein p3 (Foxp3) inhibitor. Epirubicin hydrochloride has antitumor activity.
細(xì)胞實(shí)驗(yàn)	Hep G2 cells (500 cells/well, monolayer) are plated in a 96-well plate. The next day the cells are treated with Epirubicin in the medium. At the end of the incubation periods, 15% volume of MTT dye solution is added. After 1 hr of incubation at 37℃, an equal volume of solubilization/stop solution (dimethylsul-foxide) is added to each well for an additional 1 hr incubation. The absorbance of the reaction solution at 570 nm is recorded.(Only for Reference)
體外活性	方法: 神經(jīng)膠質(zhì)瘤細(xì)胞 U-87 和神經(jīng)元原代培養(yǎng)物用 Epirubicin hydrochloride (0.5-100 μM) 處理 48 h，使用 MTT assay 檢測細(xì)胞活力。結(jié)果: Epirubicin 以濃度依賴性方式顯著降低大鼠 U-87 細(xì)胞的活力，IC50 為 6.3 μM。與 U-87 神經(jīng)膠質(zhì)瘤細(xì)胞相反，大鼠正常神經(jīng)元細(xì)胞對 Epirubicin 具有耐藥性。[1] 方法: 人乳腺癌細(xì)胞 MDA-MB-231 和 SK-BR-3 用 Epirubicin hydrochloride (2-3 μM) 處理 48 h，使用 Flow cytometry 檢測細(xì)胞凋亡。結(jié)果: Epirubicin 在 G2/M 阻滯中顯著增加了 sub-G 細(xì)胞。[2]
體內(nèi)活性	方法: 為檢測體內(nèi)抗腫瘤活性，將 Epirubicin hydrochloride (7 mg/kg) 靜脈注射給攜帶 MDA-MB-231 異種移植物的 BALB/c nu/nu 裸鼠，給藥三次，間隔四天。結(jié)果: Epirubicin 將腫瘤生長減少了一半，同時使體重減輕了 10% 以上。Epirubicin 在 7 mg/kg 時的 IR 值為 53.3%。[2]
