Chymotrypsin (Chymotrypsin A) is a serine protease produced by the pancreas. Chymotrypsin cleaves protein chains at the carboxyl side of aromatic amino acids^[1][2].

用 1 mM HCl 配制成 10 mg/mL 濃度，并添加 2 mM 氯化鈣作為穩(wěn)定劑。配制好的溶液可在 4℃ 下保存一周。

蛋白質的制備
1. 溶解及變性：將蛋白質溶解在 100mM Tris-HCl, 10mM CaCl₂ (pH 8.0) 中，難以溶解或需要變性才能有效消化的蛋白質可在室溫至 37 ℃ 下用最小體積溶解在變性劑（例如 6-8M 尿素或 6 M 鹽酸胍）中 1 h，對于某些蛋白質，將樣品加熱至 60℃ 并保持 15-20 min 可能會更有利于溶解變性；
2. 二硫化物還原：向溶解的蛋白質中添加 DTT 或β-巰基乙醇至終濃度為 5 mM。將該樣品在 50-60℃ 下加熱 20 min；
3. 烷基化：讓還原后的蛋白質混合物冷卻至室溫，添加碘乙酰胺至終濃度為 15 mM；
4. 最后用 100 mM Tris-HCl, 10mM CaCl₂ (pH 8.0) 調節(jié)反應體積，使尿素或鹽酸胍濃度低于或等于 1 M 。
酶重構
將凝乳蛋白酶溶解在 1 mM HCl 中，推薦最終濃度為 0.5-1 μg/μL。
消化
添加凝乳蛋白酶至最終蛋白酶與蛋白比例為 1：200 至 1：20 (w/w)，并在 25℃ 下孵育樣品 2-18 h。也可以通過添加 0.5% 三氟乙酸來終止消化反應。
