Uses and preparation of 3-N-Boc-aminocyclohexanone:
N-BOC-3-cyclohexanone amine is an organic intermediate that has been reported in the literature to be used in the preparation of BTK inhibitors.
The solution of 2-cyclohexen-1-one (14 ml, 150 mmol) and tert-butyl carbamate (17 g, 145.11 mmol) in DCM (30 ml) is stirred with bismuth nitrate pentahydrate (14 g, 28.8 mmol) for 21 h at room temperature. It was further diluted with DCM, filtered with hyflo, the filtrate was washed with sodium bicarbonate solution and saline, the organic phase was dried with Na2SO4, and the solvent was filtered and evaporated to obtain a crude product of 22.1 g. Silica gel chromatography (EtOAc/cyclohexanol 3:7) followed by crystallization from the same solvent system yielded N-BOC-3-cyclohexanone amine (14.43 g, 47%).
