向5.00 g (40.3 mmol) 3-氟苯甲醛中加入20 mL乙醇,隨后加入4.19 g (40.3 mmol) 丙二酸和4.66 g (60.4 mmol) 乙酸銨。在攪拌下,將混合物加熱至80至90°C,回流反應(yīng)7小時。反應(yīng)完成后,將反應(yīng)混合物冷卻至0至5°C,繼續(xù)攪拌1小時,隨后過濾,得到4.27 g 3-氨基-3-(3-氟苯基)丙酸(外消旋混合物),為白色粉末(基于3-氟苯甲醛的分離產(chǎn)率為57.9%)。 3-氨基-3-(3-氟苯基)丙酸(外消旋混合物)的物理性質(zhì)如下: 1H-NMR (δ (ppm), D2O): 2.83 (dd, 1H, J = 16.1, 6.8 Hz), 2.90 (dd, 1H, J = 16.1, 7.8 Hz), 4.67 (dd, 1H, J = 6.8, 7.8 Hz), 7.2-7.5 (m, 5H). 13C-NMR (δ (ppm), D2O): 43.4, 55.1, 116.9, 119.0, 125.7, 134.0, 141.5, 164.3, 179.9. MS (EI) m/z: 183 (M+). MS (CI, i-C4H10) m/z: 184 (MH+).
