外観
本品は白色の結(jié)晶又は結(jié)晶性の粉末である．
本品はエタノール(95)に溶けやすく，水にやや溶けにくい．
本品のエタノール(95)溶液(1→20)は旋光性を示さない．

確認(rèn)試験
(１) 本品の水溶液(1→50000)につき，紫外可視吸光度測
定法〈2.24〉により吸収スペクトルを測定し，本品のスペク
トルと本品の參照スペクトル又はグアイフェネシン標(biāo)準(zhǔn)品に
ついて同様に操作して得られたスペクトルを比較するとき，
両者のスペクトルは同一波長のところに同様の強(qiáng)度の吸収を
認(rèn)める．
(２) 本品を乾燥し，赤外吸収スペクトル測定法〈2.25〉の
臭化カリウム錠剤法により試験を行い，本品のスペクトルと
本品の參照スペクトル又は乾燥したグアイフェネシン標(biāo)準(zhǔn)品
のスペクトルを比較するとき，両者のスペクトルは同一波數(shù)のところに同様の強(qiáng)度の吸収を認(rèn)める．
定量法
本品及びグアイフェネシン標(biāo)準(zhǔn)品を乾燥し，その約
60mgずつを精密に量り，それぞれを水に溶かし，正確に
100mLとする．これらの液5mLずつを正確に量り，それぞ
れに水を加えて正確に100mLとし，試料溶液及び標(biāo)準(zhǔn)溶液
とする．これらの液につき，紫外可視吸光度測定法
により試験を行う．試料溶液及び標(biāo)準(zhǔn)溶液から得られたそれ
ぞれの液の波長273nmにおける吸光度AT及びASを測定する．
純度試験
(１) 溶狀 本品0.20gを水10mLに溶かすとき，液は無色
澄明である．
(２) 塩化物〈1.03〉 本品0.7gに水25mLを加え，加溫して
溶かし，冷後，希硝酸6mL及び水を加えて50mLとする．こ
れを検液とし，試験を行う．比較液には0.01mol/L塩酸
0.40mLを加える(0.020％以下)．
(３) 重金屬〈1.07〉 本品2.0gに水25mLを加え，加溫して
溶かし，冷後，希酢酸2mL及び水を加えて50mLとする．こ
れを検液とし，試験を行う．比較液には鉛標(biāo)準(zhǔn)液2.0mLを加
える(10ppm以下)．
(４) ヒ素〈1.11〉 本品1.0gをとり，第3法により，検液を
調(diào)製し，試験を行う(2ppm以下)．
(５) 遊離グアヤコール 本品1.0gをとり，水25mLを正確
に加え，加溫して溶かし，冷後，試料溶液とする．別にグア
ヤコール0.100gをとり，水に溶かし，正確に1000mLとする．
この液3mLを正確に量り，水22mLを正確に加え，標(biāo)準(zhǔn)溶液
とする．試料溶液及び標(biāo)準(zhǔn)溶液にヘキサシアノ鉄(Ⅲ)酸カリ
ウム試液1.0mL 及び4－ アミノアンチピリン溶液(1 →
200)5.0mLずつを加え，正確に5秒間振り混ぜる．直ちに炭
酸水素ナトリウム溶液(1→1200)を加えて正確に100mLとす
る．これらの液につき，4－アミノアンチピリン溶液を加え
たときから正確に15分後に，水25mLを用いて同様に操作し
て得た液を?qū)澱栅趣?，紫外可視吸光度測定法〈2.24〉により
試験を行うとき，波長500nmにおける試料溶液から得た液
の吸光度は，標(biāo)準(zhǔn)溶液から得た液の吸光度より大きくない．
(６) 類縁物質(zhì) 本品1.0gをエタノール(95)100mLに溶かし，
試料溶液とする．この液1mLを正確に量り，水を加えて正
確に200mLとし，標(biāo)準(zhǔn)溶液とする．これらの液につき，薄
層クロマトグラフィー〈2.03〉により試験を行う．試料溶液
及び標(biāo)準(zhǔn)溶液10μLずつを薄層クロマトグラフィー用シリカ
ゲルを用いて調(diào)製した薄層板にスポットする．次にジエチル
エーテル／エタノール(95)／アンモニア水(28)混液(40：
10：1)を展開溶媒として約10cm展開した後，薄層板を風(fēng)乾
する．これに噴霧用4－ジメチルアミノベンズアルデヒド試
液を均等に噴霧した後，110℃で10分間加熱するとき，試料
溶液から得た主スポット以外のスポットは，標(biāo)準(zhǔn)溶液から得
たスポットより濃くない．
貯法

0.5％以下(1g，減圧，60℃，3時間)．

説明
Guaifenesin is an oral expectorant drug. The expectorant action of guaifenesin is mediated by stimulation of the gastrointestinal tract. It is a common ingredient in prescription and over-the-counter medications used to treat cough due to colds and minor upper respiratory infections. Additionally, Guaifenesin is also a centrally-acting muscle relaxant and is used routinely in combination with analgesics and sedatives in large-animal veterinary surgery.
化學(xué)的特性
This substance is a white crystalline powder with a melting point of 78.5-79°C and a boiling point of 215°C (2.53kPa). It is soluble in 20ml of water and ethanol, chloroform, glycerin, and dimethylformamide. It is easily soluble in benzene but insoluble in petroleum ether. It has a slightly bitter taste and a slightly particular odor.
使用
Guaifenesin is a muscle relaxant with expectorant properties often used as an expectorant to facilitate the removal of phlegm from the airways in acute respiratory tract infections. Guaifenesin comes in tablet and capsule form, as syrup, as dissolving granules, and recently as an extended-release (longacting) tablet. The tablets, capsules, dissolving granules, and syrup are usually taken with or without food every 4 h as needed. The extended-release tablet is usually taken with or without food every 12 h.

Guaifenesin is an expectorant, widely used in the treatment of cough. Its mode of action involves the alleviating of cough discomfort by increasing sputum volume and decreasing its viscosity, thus resulting in effective cough.

Guaiphenesin facilitates secretion from bronchial mucous membranes, thus relieving a
cough in colds, bronchitis, and bronchial asthma.

Guaifenesin’s production and use as veterinary and human
medicines may result in its release to the environment through
various waste streams.
If released to air, an estimated vapor pressure of
1.5× 10–6 mm Hg at 25 ℃ indicates that guaifenesin will
exist in both the vapor and particulate phase.
Based upon an estimated Henry’s law constant of
4.4 × 10
-11 atm-m

-1, volatilization from moist soil
surfaces or from water surfaces are not expected to be important
fate processes for guaifenesin.
Guaifenesin is expected to have high mobility in soil based
upon an organic carbon–water partition coefficient (Koc) of
140, which indicates that it will have more solubility in water
and is less likely to adsorb onto organic matter in soil and
plants.
