以5-氟-1H-吲唑(2.0g,14.0mmol)為原料,將其溶解于DIVIF(50mL)中。在0℃下,向該溶液中依次加入碘(7.46g,28.0mmol)和氫氧化鉀(2.4g,42mmol)。將反應(yīng)混合物在0℃下攪拌0.5小時(shí)后,逐漸升溫至室溫。反應(yīng)進(jìn)程通過薄層色譜(TLC)進(jìn)行監(jiān)測。反應(yīng)完成后,將混合物過濾,濾液在減壓下濃縮,得到粗產(chǎn)物。粗產(chǎn)物通過硅膠柱色譜(洗脫劑:石油醚/乙酸乙酯=3/1)純化,得到5-氟-3-碘-1H-吲唑(3.7g,產(chǎn)率:96.1%),為白色固體。產(chǎn)物結(jié)構(gòu)經(jīng)1H NMR(400 MHz,DMSO-d6)確認(rèn):δ=13.64(s,1H),7.63(dd,J=4.8,4.4 Hz,1H),7.38-7.30(m,1H),7.20(dd,J=6.4,2.4Hz,1H)。
