在250 mL圓底燒瓶中加入30 mL二氯甲烷和20 mL二氯乙烷,隨后加入N,N-二甲基甲酰胺(1.8 mL,23.28 mmol)和草酰氯(2.9 mL,33.86 mmol)。在0℃下,將草酰氯緩慢滴加到混合物中。接著,加入噻吩并[3,2-b]吡啶-7(4H)-酮(1.6 g,10.58 mmol),并將反應混合物加熱回流6小時。反應完成后,將混合物冷卻至室溫,得到淡黃色固體的7-氯噻吩并[3,2-b]吡啶(1.7 g,收率90%)。產(chǎn)物經(jīng)1H-NMR(400 MHz,CDCl3)表征:δ 8.61(d,J = 5.2 Hz,1H),7.81(d,J = 5.2 Hz,1H),7.30(d,J = 4.8 Hz,1H)。
