在氬氣保護(hù)下,將N-氯代琥珀酰亞胺(0.53 g,3.95 mmol)緩慢加入至2-氨基-5-硝基吡啶(0.50 g,3.59 mmol)的無(wú)水甲苯(7.18 mL)溶液中。反應(yīng)混合物在80℃下持續(xù)攪拌1小時(shí)。反應(yīng)完成后,將混合物冷卻至室溫,加入15 mL水稀釋。隨后,用乙酸乙酯進(jìn)行萃取,依次用飽和碳酸氫鈉溶液和鹽水洗滌有機(jī)層。有機(jī)相經(jīng)無(wú)水硫酸鈉干燥后,減壓濃縮,所得粗產(chǎn)物通過(guò)柱色譜法(硅膠60N,正己烷/乙酸乙酯=80/20至60/40梯度洗脫)純化,得到黃色固體2-氨基-3-氯-5-硝基吡啶(0.40 g,2.30 mmol,收率64%)。產(chǎn)物結(jié)構(gòu)經(jīng)核磁共振氫譜(1H NMR, 500 MHz, DMSO-d6)δ 8.84 (d, J = 2.5 Hz, 1H), 8.35 (d, J = 2.5 Hz, 1H), 7.91 (brs, 1H);核磁共振碳譜(13C NMR, 125 MHz, DMSO-d6)δ 112.58, 132.14, 135.01, 145.25, 159.75;高分辨質(zhì)譜(HRMS, ESI-TOF, m/z)計(jì)算值C5H4ClN3O2Na [M + Na]+: 195.9884, 197.9856,實(shí)測(cè)值: 195.9878, 197.9847確認(rèn)。
