以5-氟喹唑啉-4(1H)-酮為原料合成4-氯-5-氟喹唑啉的一般步驟如下:將HATU(197 mg)、二異丙基乙胺(90 μL)、乙酸(22 μL)和N-[3-氯-4-(吡啶-2-基甲氧基)苯基]-5-[2-(甲氨基)乙氧基]喹唑啉-4-胺(150 mg)的DCM(20 mL)溶液攪拌2小時。反應完成后,用水洗滌反應液,再用鹽水洗滌,最后真空濃縮。使用DCM-5%甲醇作為洗脫液,通過柱色譜法純化殘余物,得到標題化合物,為白色固體(114 mg,產(chǎn)率69%)。NMR光譜(DMSO-d6)δ 1.95(s,3H),3.00(s,3H),3.89(t,2H),4.48(m,2H),5.29(s,2H),7.18(d,1H),7.24(d,1H),7.35(m,2H),7.59(m,2H),7.72(dd,1H),7.85(dt,1H),7.96(d,1H),8.46(s,1H),8.58(m,1H),9.70(bs,1H);質譜m/z 478 [M+H]+。
