以1H-吡唑并[3,4-c]吡啶為原料合成3-碘-1H-吡唑并[3,4-c]吡啶的一般步驟:將1H-吡唑并[3,4-c]吡啶(300 mg,2.52 mmol,1.00當(dāng)量)、氫氧化鉀(KOH,500 mg,8.91 mmol,3.50當(dāng)量)和二碘甲烷(1.28 g,5.04 mmol,2.00當(dāng)量)懸浮于N,N-二甲基甲酰胺(DMF,10 mL)中。反應(yīng)混合物在室溫下攪拌過夜。反應(yīng)完成后,用水淬滅反應(yīng),用乙酸乙酯萃取有機相,有機層用無水硫酸鈉干燥,減壓濃縮。粗產(chǎn)物通過硅膠柱色譜純化,洗脫劑為乙酸乙酯/石油醚(1:1,v/v),得到3-碘-1H-吡唑并[3,4-c]吡啶(577 mg,收率84%)為黃色固體。LC-MS(ESI,m/z):246 [M + H]+。
