Dihydrohomofolic acid is a substrate for dihydrofolate reductase; produces tetrahydrohomofolate which in turn inhibits thymidylate synthetase & therefore DNA synthesis.

Dihydrohomofolic acid, an analog of dihydrofolic acid (H2F), might be effective against tumor in which resistance to methotrexate (MTX) is associated with high leveIs of H2F-reductase[1].
